Powder coconut water (ACP®) as extender for semen cryopreservation of Brazilian migratory fish species

Methods to dehydrate coconut water and produce a stable and standardized powder coconut water (ACP®) has been developed. The use of ACP® as semen extender has been tested in many mammalian species, but this is the first report on fish species. The aim of this study was to compare ACP® with our standard freezing medium for semen cryopreservation of three Brazilian migratory fish species. ACP® was tested in combination with methylglycol (MG) or dimethyl sulphoxide (DMSO) and compared with our standard medium containing glucose and MG for Prochilodus lineatus and BTS® (Beltsville Thawing Solution, Minitub) and MG for Brycon orbignyanus and Leporinus obtusidens. Semen was frozen in nitrogen (N2) vapor at -170°C and then plunged into liquid N2. Highest post-thaw sperm motility was observed when semen was cryopreserved in ACP-MG and in the standard medium for all the three species. The combination of ACPDMSO was efficient only for B. orbignyanus. ACP® can be used as fish semen extender during the cryopreservation process, maintaining high post-thaw sperm motility.
